Objective: Platelet Rich Plasma (PRP) has been extensively utilized as a therapy for many musculoskeletal diseases. Despite its broad use clinically, there is still an unmet need in standardizing PRP formulations, specifically among two main formulations, Leukocyte Rich PRP (LR-PRP) and Leukocyte Poor PRP (LP-PRP). One critical step towards standardizing PRP is to comprehensively characterize participating cell types in PRP, namely with respect to leukocyte composition.
Design: Prospective Human Ex-Vivo AnalysisSetting : Controlled Laboratory StudyParticipants : 12 healthy adult males between ages 25 to 31

Main Outcome Measures: Leukocyte Profile in Human LR-PRP
Results: In LR-PRP, lymphocytes were the predominant WBC type (11.94 ± 2.97 x 103 cells/μL) followed by neutrophils (3.72 ± 1.28 x 103 cells/μL). The mean cumulative percentage of granulocytes was 25% ± 8% and agranulocytes was 75% ± 9%. There was a significant difference observed between granulocyte and agranulocyte percentage within both WB and LR-PRP groups. In addition, there was a significant difference observed between the WB and LR-PRP granulocyte percentages and between the WB and LR-PRP agranulocyte percentages.Conclusions: LR-PRP is predominantly lymphocyte rich but contains other WBC sub-types that are not currently reported which could affect LR-PRP’s therapeutic efficacy.
