Objective: The aim of this study was to test the hypothesis that O-GlcNAcylation plays a protective role by reducing cerebral hemorrhage volume, reducing cerebral edema, and improving neurological function score in cerebral hemorrhage.
Design: basic research Setting : Stroke rehabilitation hospital Participants : Male C57/BL6 mice, 6-8 weeks
Interventions: The model of cerebral hemorrhage was established by stereotactic localization autologous blood injection .MNSS score, foot wrong step experiment, corner experiment and forepaw placement experiment were used to test whether O-GlcNAcylation had a protective effect on nerve function, the effect of O-GlcNAcylation on hematoma and edema in brain tissue was detected by pathology.
Main Outcome Measures: The effect of glycosylation on cerebral hemorrhage was detected by pathology, neurological function score and A method of measuring wet and dry weight.
Results: First, the intracerebral hemorrhage model of mice was established by autologous blood injection, which was divided into two groups: the upregulated and down-regulated O-glcnac glycosylation group.After 24 hours of modeling, the volume of cerebral hematoma in the group o-glcnac glycosylation up-regulated was decreased, while the volume of cerebral hemorrhage in the group down-regulated glycosylation was significantly increased. The neurological function of the mice was evaluated 24 and 72 hours after the onset of intracerebral hemorrhage.O-glcnac glycosylation increased, and the neurological function score of corner experiment significantly improved, and also o-glcnac glycosylation group’s mNSS neurological function score in intracerebral hemorrhage was improved.We observed that the neurological function score of mice in the group of increased o-glcnac glycosylation was improved in the experiment.O-glcnac glycosylation improves nerve function in intracerebral hemorrhage. The degree of brain edema was significantly reduced in the o-glcnac glycosylation up-regulated group. Conclusions: This study will reveal the protective effect of o-glcnac glycosylation on intracerebral hemorrhage, clarify its protective effect, provide experimental basis and theoretical basis for clinical treatment of intracerebral hemorrhage.
